Description

This Sandwich ELISA Kit from Assay Genie provides a quantitative immunoassay for the detection of GSK-3 Beta (Glycogen Synthase Kinase 3 Beta) in human samples. It is configured for reactivity with human specimens and is intended for cell biology research, supporting investigation of GSK-3 Beta—a key enzyme in processes including cell growth, differentiation, and apoptosis—and its reported relevance to diseases such as Alzheimer's disease, cancer, and diabetes.

The double-antibody sandwich ELISA uses a colorimetric detection method and is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Reported sensitivity is 0.375 ng/mL, with a measuring range of 0.625–40 ng/mL. Linearity in serum is given as 85–97% (1:2), 83–101% (1:4) and 82–100% (1:8); EDTA plasma as 88–102%, 83–100% and 81–95%; heparin plasma as 95–104%, 83–96% and 80–100%. Recovery averages 94% in serum (range 87–103%), 99% in EDTA plasma (range 97–101%), and 93% in heparin plasma (range 86–105%). The procedure comprises reagent equilibration, primary incubation at 37 °C for 90 minutes, biotinylated detection antibody binding for 60 minutes, HRP-streptavidin incubation for 30 minutes, TMB colour development in the dark for 10–20 minutes, and absorbance measurement at 450 nm after adding stop solution.

Reagents are stored at 2–8 °C for up to 12 months. The kit includes a dismountable ELISA microplate, lyophilised standard, biotin-labelled antibody concentrate, HRP-streptavidin conjugate (SABC) concentrate, TMB substrate, sample dilution buffer, antibody dilution buffer, SABC dilution buffer, stop solution, wash buffer concentrate, plate sealers, and a technical manual. Sample preparation is described for serum (clot, centrifuge at 1000 × g for 20 minutes), plasma (anticoagulant tubes, centrifuge at 1000 × g for 15 minutes at 2–8 °C), tissue homogenate (PBS with protease inhibitors), cell culture supernatant (centrifuge at 2500 rpm for 5 minutes), and cell lysate (lysis buffer with protease inhibitors). Additional support for other sample types is directed to the manufacturer's technical team.
