GRB7 is an SH2-domain adaptor protein that binds phosphotyrosine motifs on activated receptors, including HER2/ErbB2. Co-amplification of GRB7 with HER2 has been documented in a subset of breast cancers, and elevated GRB7 expression has been reported in esophageal and pancreatic tumor tissues. The protein participates in pathways governing cell migration, proliferation, and angiogenesis through downstream effectors such as focal adhesion kinase (FAK) and ERK1/2.
