GRAP is encoded by the human GRAP gene (NCBI Gene ID 10750; UniProt Q13588; OMIM 604330), producing a 217-residue cytoplasmic adaptor of approximately 25-27 kDa that is most highly expressed in thymus and spleen tissue. The protein's SH2 domain binds ligand-activated receptor tyrosine kinases such as c-Kit and the erythropoietin receptor, and forms a stable complex with the BCR-ABL oncoprotein, while its N-terminal SH3 domain recruits the Ras guanine nucleotide exchange factor SOS1, linking proximal kinase activity to downstream Ras/MAPK signaling relevant to leukemia and hematopoietic research.
The Assay Genie workflow uses a 100 µL sample loading volume with a 90 minute primary incubation at 37 °C, followed by a biotinylated detection antibody (60 minute binding at 37 °C), HRP-Streptavidin conjugate (30 minute binding at 37 °C), and TMB chromogen development in the dark before stop solution is added. Reconstituted standard vials should be aliquoted and frozen at -20 °C to limit repeated freeze-thaw cycles, and sealed kit components remain stable when refrigerated.
