Description

A Sandwich ELISA developed by Assay Genie for the quantitative measurement of human Granzyme M (GZMM). The assay is intended for use on serum, plasma, tissue homogenates, cell culture supernates and other biological fluids, and shows reactivity with human samples. Target synonyms recognised by the kit include Met-1 serine protease, Met-ase, Natural killer cell granular protease, Hu-Met-1 and MET1.

Performance specifications reported by the manufacturer include a detection range of 78–5000 pg/mL, a sensitivity of 20 pg/mL, an intra-assay CV of 5.8% and an inter-assay CV of 8.5%. Linearity has been evaluated at dilution factors of 1:2, 1:4, 1:8 and 1:16 in serum, EDTA plasma and heparin plasma. Recovery in serum averages 103% (range 97–109%) and in plasma averages 105% (range 99–111%). Specificity has been confirmed against natural and recombinant human Granzyme M (UniProt P51124).

Granzyme M is a secreted serine protease of the granzyme subfamily that cleaves peptide substrates after methionine, leucine and norleucine, with reported physiological substrates including EZR, alpha-tubulins and the apoptosis inhibitor BIRC5/Survivin. It promotes caspase activation and apoptosis in target cells and is implicated in immune responses linked to cancer, autoimmune disorders and viral infections. The assay is read colourimetrically at 450 nm. For research use only.
