Description

A sandwich ELISA-based colorimetric kit from Assay Genie is designed for the in vitro quantitative determination of human Granzyme B (GzmB) in serum, plasma and other biological fluids. The assay uses a double-antibody sandwich format with a reported sensitivity of 0.938 pg/ml and a measurable range of 1.563–100 pg/ml. Reactivity is specific to human samples.

Performance characteristics include intra-assay CV below 8% and inter-assay CV below 10%. Recovery has been validated in serum (85–104%, average 95%), EDTA plasma (85–96%, average 91%) and UFH plasma (88–99%, average 91%), and linearity has been demonstrated by serial dilution across the same matrices. The kit components include a dismountable microplate, lyophilized standard, biotin-labelled antibody, HRP-streptavidin conjugate (SABC), TMB substrate, stop solution and wash buffer, with most components stored at 4°C for up to six months. Sample preparation guidance covers serum, plasma (EDTA or heparin), urine, cerebrospinal fluid, cell culture supernatants, cell lysates, tissue homogenates and breast milk, with centrifugation and −80°C storage as appropriate.

Granzyme B (UniProt P10144) is a serine protease of the granzyme subfamily, secreted by natural killer cells and cytotoxic T lymphocytes and proteolytically processed to an active form that induces target cell apoptosis through cleavage of caspases-3, -7, -9 and -10. The gene maps to chromosomal location 14q12, and the protein localises to the cytoplasm, cytosol, immunological synapse, intracellular membrane-bound organelles, membrane and nucleus. It is implicated in apoptosis, natural killer cell mediated cytotoxicity and protein processing, and the kit supports studies in immunology, oncology and infectious disease research. The product is for research use only.
