The GPBAR1 protein, also known as TGR5 or M-BAR, is a membrane-bound G protein-coupled receptor activated by primary and secondary bile acids such as lithocholic acid and deoxycholic acid. It regulates energy expenditure, glucose homeostasis, and inflammatory signaling, and is expressed in brown adipose tissue, skeletal muscle, enteroendocrine L-cells, and biliary epithelial cells. Researchers studying metabolic syndrome, type 2 diabetes, non-alcoholic fatty liver disease, and bile acid signaling pathways commonly use this ELISA to quantify circulating GPBAR1 as a biomarker of receptor activity.

Each kit from Assay Genie is supplied as a complete immunoassay in a microplate format, with pre-coated wells, biotinylated detection antibody, lyophilized standards, and substrate reagents. Store the kit refrigerated; the sealed components remain stable through the printed expiration date. Bring all reagents to room temperature before use, and avoid repeated freeze-thaw cycles of the reconstituted standards by aliquoting into single-use vials.

For assay setup, dilute samples with the provided assay buffer according to the enclosed protocol, then run the substrate reaction and read absorbance in a standard plate reader at the recommended wavelength. A sample volume of 100 uL per well is typical, and the kit is calibrated against a recombinant human GPBAR1 standard curve for accurate quantification. The kit is intended for research use only and is not for diagnostic or clinical procedures.
