Human Golgi Protein 73/GP-73 ELISA Kit

From Assay Genie, this Sandwich ELISA Kit is designed for the quantitative measurement of human Golgi Protein 73 (GP-73), a resident protein of the Golgi apparatus that has been studied as a biomarker in liver disease research, including hepatitis and liver cancer.

Using a colorimetric, double-antibody sandwich format, the assay has a sensitivity of 1.875 ng/mL and a working range of 3.125–200 ng/mL. It is validated for use with human serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid sample types, with components intended to be stored at 2–8°C for 12 months. The protocol uses a 90-minute primary incubation at 37°C, a 60-minute biotin-labelled detection antibody step, 30 minutes of HRP-Streptavidin (SABC) binding, 10–20 minutes of TMB substrate development in the dark, and absorbance reading at 450 nm after addition of stop solution.

Validation data from the manufacturer report linearity at 1:2, 1:4 and 1:8 dilutions of 86–102%, 87–100% and 90–99% for serum (n = 5); 85–98%, 82–97% and 83–95% for EDTA plasma (n = 5); and 85–102%, 88–100% and 84–98% for heparin plasma (n = 5). Recovery averages are reported as 92% for serum (range 85–102%), 96% for EDTA plasma (range 92–99%) and 98% for heparin plasma (range 86–103%).
