Human GOAT/MBOAT4 ELISA Kit

This Sandwich ELISA kit from Assay Genie is designed for the quantitative detection of human GOAT/MBOAT4 (ghrelin O-acyltransferase / membrane-bound O-acyltransferase domain-containing protein 4, UniProt Q96T53). The assay uses a colorimetric detection method in a double-antibody sandwich format and is validated for use with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples.

The kit reports a sensitivity of 0.188 ng/mL and a working range of 0.313–20 ng/mL. Reported linearity (n = 5) across dilution ratios of 1:2, 1:4 and 1:8 is 87–102%, 87–94% and 84–101% for serum; 84–96%, 83–99% and 82–97% for EDTA plasma; and 83–89%, 82–96% and 85–96% for heparin plasma. Spike-and-recovery performance is 85–105% (average 91%) for serum, 86–100% (average 95%) for EDTA plasma, and 88–103% (average 95%) for heparin plasma.

MBOAT4 is described as a key enzyme in lipid metabolism and the biosynthesis of phosphatidylinositol, a key constituent of cell membranes, and its dysregulation has been linked to cancer, metabolic disorders and neurodegenerative conditions. The assay protocol involves equilibrating reagents and TMB substrate to room temperature, primary incubation at 37°C for 90 minutes, biotinylated detection antibody incubation at 37°C for 60 minutes, HRP-streptavidin binding at 37°C for 30 minutes, TMB colour development in the dark for 10–20 minutes, and absorbance measurement at 450 nm after addition of stop solution. Kit components should be stored at 2–8°C for up to 12 months.
