Human GNAQ ELISA Kit (HUFI04090)

The Human GNAQ ELISA Kit from Assay Genie is built for quantitative measurement of the alpha subunit of heterotrimeric G-protein Gq, a 42 kDa GTP-binding signaling protein encoded by the GNAQ gene on chromosome 9q21. This target couples G-protein-coupled receptors to phospholipase C-beta, mobilizing intracellular calcium and activating protein kinase C pathways downstream of receptors for thrombin, endothelin, and various neurotransmitters.

Researchers commonly apply this assay to investigate GNAQ protein levels in patient samples collected from individuals with uveal melanoma, where activating somatic mutations (notably Q209L and Q209P) constitutively drive MAPK signaling, as well as in Sturge-Weber syndrome and port-wine stain birthmarks, both linked to mosaic GNAQ R183Q variants. It is also useful in cardiovascular pharmacology studies probing Gq-coupled receptor desensitization and in cell line authentication workflows that confirm wild-type or mutant GNAQ expression in engineered HEK293 and melanocyte models.

Each microplate-format kit is supplied with a pre-coated plate containing removable strip wells, biotin-conjugated detection reagent, streptavidin-HRP working solution, calibrated recombinant standards, wash concentrate, TMB substrate, stop reagent, and adhesive plate sealers. Standard curves are constructed from a serial dilution of the included calibrator, and absorbance is read on a standard microplate reader. Reconstituted standards and unused strips should be aliquoted to limit freeze-thaw cycles and stored refrigerated until the indicated expiration date.

The recommended sample volume is 100 µL per well, and samples should be clarified by centrifugation at 1,000 x g for 10 minutes prior to dilution. This product is intended for research use only and is not for diagnostic or therapeutic procedures.
