Description

This Sandwich ELISA kit from Assay Genie is designed for the quantitative detection of human Glycogen phosphorylase, liver form (PYGL) in serum, plasma, tissue homogenates, cell culture supernates and other biological fluids. The liver isoform of glycogen phosphorylase is an allosteric enzyme central to carbohydrate metabolism, catalysing the cleavage of alpha-1,4-glucosidic bonds to release glucose-1-phosphate from hepatic glycogen stores and thereby supporting glucose homeostasis.

The assay has a stated detection range of 0.312–20 ng/mL and a sensitivity of 0.156 ng/mL. Reported precision is an intra-assay CV of 7.8% and an inter-assay CV of 9.1%. Linearity has been assessed in serum, EDTA plasma and heparin plasma across dilutions of 1:2, 1:4, 1:8 and 1:16, with serum values spanning 85–113% across dilutions and plasma values spanning 80–119%. Recovery is reported at an average of 101% (range 95–107%) for serum and 103% (range 97–109%) for plasma. Specificity covers natural and recombinant human Glycogen phosphorylase, liver form.

Dysregulation of PYGL is relevant to metabolic disease: defects in the gene cause glycogen storage disease type 6 (GSD6, also known as Hers disease), a condition characterised by mild to moderate hypoglycaemia, mild ketosis, growth retardation and prominent hepatomegaly. The kit is therefore suited to research into glycogen metabolism, glucose homeostasis and related metabolic disorders, and is supplied for research use only.
