Description

Assay Genie provides a colorimetric sandwich ELISA designed for the quantitative detection of human Glutathione S-transferase theta-2 (GSTT2) in serum, plasma, tissue homogenates, cell culture supernates and other biological fluids. The assay targets natural and recombinant human GSTT2 and is intended for research use, supporting studies where the enzyme serves as a biomarker of cellular detoxification, including investigations of oxidative stress, environmental exposures and cancer development.

GSTT2 is a cytoplasmic homodimer (UniProt P0CG29) belonging to the GST superfamily, Theta family (EC 2.5.1.18), with a molecular weight of 27,506 Da and a chromosomal locus at 22q11.23 (NCBI Gene ID 2953). It catalyses the conjugation of reduced glutathione to a broad range of exogenous and endogenous hydrophobic electrophiles and also displays sulfatase activity, consistent with a role in xenobiotic metabolism and glutathione-related biochemistry.

The assay has a detection range of 0.312–20 ng/mL and a sensitivity of 0.173 ng/mL. Intra-assay CV is 6.8% and inter-assay CV is 11.5%. Dilutional linearity for serum (N=5) is 108–118% at 1:2, 101–111% at 1:4, 88–98% at 1:8 and 85–97% at 1:16; for EDTA plasma (N=5) it is 102–111%, 90–99%, 106–115% and 87–95%; and for heparin plasma (N=5) it is 113–123%, 84–84%, 91–103% and 103–113%. Spike-and-recovery averages 99% (range 93–105%) in serum and 101% (range 95–107%) in plasma.
