Human Glutathione Peroxidase 2/GPX2 ELISA Kit

From Assay Genie, this Sandwich ELISA kit is designed for the quantitative measurement of human Glutathione Peroxidase 2 (GPX2) in serum, plasma, cell culture supernatants, cell or tissue lysates and other liquid samples. The assay uses a colorimetric detection method in a double antibody sandwich format and is intended for research applications related to oxidative stress and antioxidant defence mechanisms, with relevance to diseases such as cancer, inflammatory disorders and neurodegenerative conditions.

The manufacturer reports a sensitivity of 46.875 pg/mL and a detection range of 78.125–5000 pg/mL. Linearity across dilutions of 1:2, 1:4 and 1:8 (n=5) is reported as 88–105%, 99–104% and 91–101% for serum; 89–98%, 85–99% and 83–100% for EDTA plasma; and 80–100%, 80–94% and 81–99% for heparin plasma. Recovery averages are 95% for serum (range 89–104%), 96% for EDTA plasma (range 87–104%) and 94% for heparin plasma (range 85–103%).

The assay procedure involves reagent and plate preparation, primary incubation at 37 °C for 90 minutes, biotin-labelled detection antibody incubation at 37 °C for 60 minutes, HRP-Streptavidin binding for 30 minutes, TMB substrate incubation in the dark for 10–20 minutes, and absorbance measurement at 450 nm after addition of stop solution. Components supplied include a pre-coated dismountable ELISA microplate, lyophilised standard, 100× biotin-labelled antibody concentrate, 100× HRP-Streptavidin conjugate, TMB substrate, sample dilution buffer, antibody dilution buffer, SABC dilution buffer, stop solution, 25× wash buffer and a plate sealer. The kit is stored at 2–8 °C for up to 12 months.
