Human Glucosidase 2 subunit beta/PRKCSH ELISA Kit

A sandwich ELISA kit from Assay Genie designed for the quantitative detection of Glucosidase 2 subunit beta (PRKCSH) in human samples. The assay uses a double-antibody sandwich format with colorimetric readout, and is intended for research in cell biology, particularly studies of protein folding and N-linked glycosylation, where PRKCSH dysregulation has been linked to cancer, diabetes and neurodegenerative disorders. The standard protocol involves equilibrating reagents and TMB substrate to room temperature; a primary incubation at 37°C for 90 minutes to allow antigen binding; biotin-labeled detection antibody incubation at 37°C for 60 minutes; HRP-Streptavidin (SABC) binding at 37°C for 30 minutes; TMB color development in the dark for 10–20 minutes; and a stop step with absorbance measured at 450 nm. Reagents are stored at 2-8°C for 12 months.

Reported linearity across serum, EDTA plasma and heparin plasma at dilutions of 1:2, 1:4 and 1:8 falls within 81-104%. Recovery ranges span 86-104% with averages between 95-100% across the same sample types. Sample preparation guidance includes centrifuging clotted blood at 1000 × g for 20 minutes for serum, anticoagulant tubes centrifuged at 1000 × g for 15 minutes at 2-8°C for plasma, homogenisation in PBS with protease inhibitors for tissue, centrifugation at 2500 rpm for 5 minutes for cell culture supernatant, and lysis buffer with protease inhibitors for cell lysates.
