Human GLS/Glutaminase kidney isoform, mitochondrial ELISA Kit

GLS is an important enzyme that plays a crucial role in glutamine metabolism, impacting processes such as cell growth, energy production, and regulation of cellular signalling pathways. Dysregulation of GLS has been implicated in various diseases including cancer, metabolic disorders, and neurodegenerative conditions, making it a valuable biomarker for studying these pathologies and developing potential therapeutic interventions. Performance characteristics include a sensitivity of 0.188 ng/mL and a detection range of 0.313–20 ng/mL. Linearity at 1:2, 1:4 and 1:8 dilutions spans 88–104%, 90–105% and 86–105% for serum (n=5); 82–99%, 90–99% and 87–101% for EDTA plasma (n=5); and 82–100%, 84–99% and 82–99% for heparin plasma (n=5). Recovery averages 94% across serum (range 85–102%), EDTA plasma (86–102%) and heparin plasma (86–103%). The procedure covers reagent and plate preparation, primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody binding at 37°C for 60 minutes, HRP-Streptavidin (SABC) binding at 37°C for 30 minutes, TMB colour development in the dark for 10–20 minutes, and a stop reaction with absorbance read at 450 nm. Sample preparation guidance is provided for serum, plasma, tissue homogenate, cell culture supernatant and cell lysate. The kit includes an ELISA microplate, lyophilised standard, biotin-labelled antibody (100X), HRP-Streptavidin conjugate (SABC, 100X), TMB substrate, sample, antibody and SABC dilution buffers, stop solution, wash buffer (25X), plate sealer and a technical manual.
