Description

From Assay Genie, this is a Sandwich ELISA, Double Antibody kit designed to measure human GLA (Alpha-galactosidase A). It is intended for use with human samples in cell biology research, and is compatible with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid sample types.

The assay uses a colorimetric detection method, with a sensitivity of 46.875 pg/mL and a measurable range of 78.125–5000 pg/mL. Reagents are stored at 2–8°C for up to 12 months. The procedure follows a standard sandwich ELISA workflow: reagent and plate preparation, primary incubation at 37°C for 90 minutes to allow antigen binding, biotin-labelled detection antibody incubation at 37°C for 60 minutes, HRP-Streptavidin binding at 37°C for 30 minutes, TMB colour development in the dark for 10–20 minutes, and absorbance reading at 450 nm after the stop solution.

Alpha-galactosidase A is an enzyme involved in breaking down complex sugars in the body, and deficiencies in this enzyme can lead to conditions such as Fabry disease. Reported linearity values (n = 5) are 89–96% at 1:2, 83–97% at 1:4, and 85–98% at 1:8 for serum; 85–95% / 82–98% / 80–96% for EDTA plasma; and 88–105% / 84–101% / 82–99% for heparin plasma. Reported recovery values (n = 5) average 99% for serum (range 95–103%), 94% for EDTA plasma (range 85–104%), and 93% for heparin plasma (range 85–105%).
