Gelsolin (synonyms: AGEL, Actin-depolymerizing factor, Brevin, ADF) is a calcium-regulated, actin-modulating protein that binds to the plus (or barbed) ends of actin monomers or filaments, preventing monomer exchange (end-blocking or capping). It can promote the assembly of monomers into filaments (nucleation) as well as sever filaments already formed, and plays a role in ciliogenesis. The protein is implicated in cellular processes such as cell motility, adhesion, and migration, and dysregulation has been linked to various diseases including cancer, Alzheimer's disease, and inflammatory conditions.
