From Assay Genie, this in vitro quantitative immunoassay measures General Control Non-depressible 5 (GCN5), also known as KAT2A, a lysine acetyltransferase that modifies histone H3 lysine residues and operates within the SAGA transcriptional coactivator complex. The dual-antibody format pairs a pre-coated capture antibody with a biotinylated detection reagent and an HRP-streptavidin conjugate, producing a chromogenic readout proportional to human GCN5 binding in each well.

The protocol uses a 100 µL sample volume per well, beginning with a 90 minute incubation at 37 °C, followed by a 60 minute incubation at 37 °C after streptavidin-HRP addition. TMB substrate reacts for 10 to 20 minutes in the dark, the reaction is stopped with the supplied stop solution, and absorbance is read on a standard microplate reader. Reagents, including the standard solution and 25X wash buffer concentrate, should be stored refrigerated and remain stable through the kit's shelf life.

Specificity testing has shown no significant cross-reactivity or interference between K-Acetyltransferase 2A (KAT2A) and analogues, supporting accurate quantitation of human GCN5 in serum-type, plasma-type, and cell lysate preparations. Researchers apply this kit to studies of altered histone acetylation in cancer models, neurodegenerative disease, metabolic disorders, and DNA damage response where KAT2A activity is dysregulated. Intended for research use only.
