Description

From Assay Genie, this Sandwich ELISA kit is designed for the quantitative measurement of gastric intrinsic factor in human samples. The kit uses a double-antibody sandwich ELISA format with colorimetric detection, and is reactive with human samples including serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples.

Reported performance characteristics include a sensitivity of 1.875 ng/mL and an assay range of 3.125–200 ng/mL. Storage is specified at 2–8°C for 12 months. Linear dilution performance is reported for serum (98–104% at 1:2; 85–103% at 1:4; 86–103% at 1:8), EDTA plasma (84–100%; 83–94%; 85–96%) and heparin plasma (80–95%; 81–99%; 88–95%). Spike-and-recovery ranges are reported as serum 95–105% (average 94%), EDTA plasma 86–101% (average 96%) and heparin plasma 92–103% (average 93%). The associated UniProt entry is P27352, with listed synonyms including GIF, IF, IFMH, INF and TCN3.

The assay workflow involves equilibrating reagents and TMB substrate to room temperature, a 90-minute primary incubation at 37°C, a 60-minute biotin-labelled detection antibody step, a 30-minute HRP-Streptavidin (SABC) binding step, 10–20 minutes of colour development with TMB substrate in the dark, and a stop reaction followed by absorbance reading at 450 nm. Gastric intrinsic factor is described as a key protein involved in the absorption of vitamin B12 in the stomach, with deficiencies linked to conditions such as pernicious anemia, making accurate measurement relevant to research in gastroenterology, nutrition and vitamin deficiency management.
