Description

From Assay Genie, this Sandwich ELISA Kit is designed for the quantitative detection of human Gamma-secretase-activating protein (PION), also known as Protein pigeon homolog or GSAP, in human samples. The kit is intended for research use only and supports work in the neurosciences area, where PION's role as a regulator of gamma-secretase activity is relevant to amyloid-beta production and conditions such as Alzheimer's disease.

According to the manufacturer, the assay has a detection range of 1.56-100ng/mL and a sensitivity of 0.78ng/mL. Reported precision values are an intra-assay CV of 6.5% and an inter-assay CV of 11.6%. Linearity has been assessed in serum (85-116% across dilutions), EDTA plasma (93-117%) and heparin plasma (86-119%), with average recovery of 111% (105-117%) for serum and 113% (107-119%) for plasma. The kit is validated for serum, plasma, tissue homogenates, cell culture supernatants and other biological fluids, and shows specificity for natural and recombinant human Gamma-secretase-activating protein.

The target protein (UniProt A4D1B5, gene GSAP, NCBI Gene ID 54103) is a Golgi apparatus and trans-Golgi network-localised activator that interacts with APP and the gamma-secretase complex, with a reported molecular weight of 28,563 Da and chromosomal location 7q11.23. Supplied components include a lyophilised standard, sample and assay diluents, detection reagents, wash buffer, substrate, stop solution and plate sealer, with storage temperatures varying by component.
