Description

This Sandwich ELISA from Assay Genie is designed for the quantitative detection of human Gamma-glutamyltranspeptidase 1 (GGT1), a membrane-anchored enzyme involved in extracellular glutathione breakdown and cellular antioxidant defence. The assay is validated for use with human serum, plasma, tissue homogenates, cell culture supernates and other biological fluids, with specificity for natural and recombinant human Glutathione hydrolase 1 proenzyme.

Performance characteristics include a detection range of 0.78–50 ng/mL, a sensitivity of 0.22 ng/mL, an intra-assay CV of 6.1% and an inter-assay CV of 9.2%. Linearity was assessed at dilutions of 1:2, 1:4, 1:8 and 1:16, with ranges in serum of 104–113%, 92–100%, 98–110% and 82–94% respectively; in EDTA plasma of 97–105%, 87–97%, 98–107% and 108–117%; and in heparin plasma of 98–107%, 104–113%, 93–104% and 109–119%. Average recovery was 102% (range 96–108%) in serum and 104% (range 98–110%) in plasma.

GGT1 (UniProt P19440) is a heterodimer of light and heavy chains anchored to the external face of the plasma membrane, with the active site located in the light chain. The enzyme initiates glutathione catabolism to release free glutamate and supply cysteine for intracellular GSH maintenance, and is implicated in xenobiotic metabolism and inflammatory regulation. Abnormal GGT1 levels have been associated with liver disease, cardiovascular disease and diabetes, supporting its use as a biomarker in cell biology research.
