Built for cardiovascular, renal, and hematology research programs, this colorimetric immunoassay from Assay Genie delivers quantitative measurement of Gamma adducin (ADD3), the membrane-skeletal protein that caps fast-growing actin filaments and links the spectrin–actin lattice to the plasma membrane. The kit is supplied as a complete 96-well microplate pack containing pre-coated strip wells, lyophilized recombinant standard, biotinylated detection antibody, HRP-streptavidin conjugate, TMB substrate, stop solution, wash and dilution buffers, plate sealer, and a technical manual, so a laboratory can move straight into the sandwich ELISA workflow without additional reagent sourcing.

For specimen handling, serum is collected by allowing blood to clot and centrifuging at 1000 x g for 20 minutes; plasma tubes are spun at 1000 x g for 15 minutes at 2–8 degrees C, while cell-culture supernatant is clarified at 2500 rpm for 5 minutes before loading. The assay has been validated against human ADD3 (UniProt Q9UEY8) with no significant cross-reactivity reported for the related alpha and beta adducin subunits, supporting its use in studies of salt-sensitive hypertension, erythrocyte membrane mechanics, and renal sodium handling.

Reagent components are stable at 2–8 degrees C for up to 12 months, with lyophilized standard and coated strips held at –20 degrees C for long-term storage and protected from light for the biotinylated antibody, SABC, and TMB working solutions. The recommended run sequence is 90 minutes at 37 degrees C for sample binding, 60 minutes for detection antibody, 30 minutes for HRP-streptavidin, and 10–20 minutes of TMB development in the dark, followed by absorbance readout.
