Human FZD4/Frizzled 4 ELISA Kit

Designed for the accurate quantification of Frizzled 4 in human research samples, this Sandwich ELISA from Assay Genie uses a colorimetric detection workflow suited to studies of the Wnt signaling pathway. Frizzled 4 is a key receptor protein implicated in cell proliferation, differentiation, and tissue development, with dysregulation associated with cancer, developmental disorders, and neurodegenerative conditions. The kit accepts serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, and is intended for storage at 2-8°C for 12 months.

Reported sensitivity is 0.469 ng/mL with an analytical range of 0.781-50 ng/mL. Linearity in serum (n=5) is 88-101% at 1:2, 90-97% at 1:4, and 85-97% at 1:8; EDTA plasma (n=5) shows 89-99%, 83-101%, and 81-100% respectively; heparin plasma (n=5) shows 90-104%, 84-98%, and 85-93%. Recovery averages 99% in serum (n=5, range 90-104%), 100% in EDTA plasma (n=5, range 95-102%), and 96% in heparin plasma (n=5, range 88-105%).

The workflow involves equilibrating reagents and TMB substrate to room temperature, primary incubation at 37°C for 90 minutes, biotinylated detection antibody binding at 37°C for 60 minutes, HRP-Streptavidin binding at 37°C for 30 minutes, color development with TMB substrate in the dark for 10-20 minutes, and a stop reaction with absorbance measurement at 450 nm.
