Description

From Assay Genie, this Sandwich ELISA kit is designed for the quantitative measurement of human Filamin C, a protein that plays a crucial role in maintaining the structural integrity of cells, especially in muscle tissues. The assay uses a double-antibody sandwich format with HRP-Streptavidin (SABC) detection and TMB substrate, with absorbance read at 450 nm. The kit is reactive with human samples and is validated for serum, plasma, cell culture supernatant, cell or tissue lysates, and other liquid samples. Reported sensitivity is 0.188 ng/mL with a standard range of 0.313–20 ng/mL. Components should be stored at 2–8°C for 12 months.

Linearity has been evaluated at 1:2, 1:4 and 1:8 dilutions: serum (n=5) 94–105%, 91–101% and 88–105%; EDTA plasma (n=5) 82–97%, 85–101% and 85–97%; heparin plasma (n=5) 80–97%, 84–99% and 82–96%. Recovery averages 99% for serum (n=5, range 91–105%), 95% for EDTA plasma (n=5, range 87–103%) and 95% for heparin plasma (n=5, range 86–101%). The assay procedure includes equilibrating reagents and the TMB substrate to room temperature, primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody incubation for 60 minutes, HRP-Streptavidin binding for 30 minutes, and TMB color development in the dark for 10–20 minutes, followed by stop solution and immediate absorbance measurement.

Dysregulation of filamin C has been linked to various muscular disorders, making it a key biomarker for studying these conditions and exploring therapeutic interventions in cell biology research.
