Human FGF 23/Fibroblast Growth Factor 23 ELISA Kit

This sandwich ELISA from Assay Genie is designed to quantify human FGF-23 in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, using a double-antibody format with colorimetric detection. The assay has a sensitivity of 9.375 pg/mL and a measuring range of 15.625–1000 pg/mL, with all kit components stable for 12 months when stored at 2–8°C.

The six-step protocol begins with reagent and TMB substrate equilibration to room temperature, followed by primary incubation at 37°C for 90 minutes, biotin-labeled detection antibody binding at 37°C for 60 minutes, HRP-Streptavidin (SABC) binding at 37°C for 30 minutes, TMB color development in the dark for 10–20 minutes, and absorbance measurement at 450 nm after addition of stop solution. Validated linearity for serum (n = 5) at 1:2, 1:4, and 1:8 dilutions is 86–102%, 86–98%, and 85–101%; for EDTA plasma (n = 5) 82–96%, 83–97%, and 89–105%; and for heparin plasma (n = 5) 86–99%, 82–90%, and 87–96%. Recovery ranges are 86–99% (average 95%) for serum, 87–101% (average 95%) for EDTA plasma, and 90–99% (average 93%) for heparin plasma.

FGF-23 is a regulator of phosphate and vitamin D metabolism linked to bone health, cardiovascular function, chronic kidney disease, and mineral disorders, with the kit intended for developmental biology research.
