Human Ferritin ELISA Kit

From Assay Genie, this Sandwich ELISA kit uses a double-antibody format with TMB-based colorimetric detection to quantify human ferritin, with absorbance read at 450 nm after stop solution is added. Ferritin is a key protein involved in iron storage and regulation, and is used as a biomarker in research on iron deficiency anemia, hemochromatosis and inflammatory disorders, supporting studies of iron metabolism and related disease processes.

The kit is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Linearity for serum (n=5) is 82-100% at 1:2, 85-100% at 1:4 and 89-100% at 1:8; EDTA plasma (n=5) 81-98%, 86-98% and 82-101%; heparin plasma (n=5) 85-102%, 86-98% and 82-94%. Recovery is 86-104% (average 92%) for serum, 89-96% (93%) for EDTA plasma and 91-102% (96%) for heparin plasma (all n=5). Components include a dismountable ELISA microplate, lyophilized standard, biotin-labeled antibody (100X), HRP-Streptavidin conjugate (100X), TMB substrate, sample/antibody/SABC dilution buffers, stop solution, 25X wash buffer, plate sealer and technical manual.

The procedure equilibrates reagents and TMB substrate to room temperature before loading standards, samples and controls for a 37°C primary incubation of 90 minutes, followed by a 60-minute biotin-labeled detection antibody incubation, a 30-minute HRP-Streptavidin (SABC) incubation, 10-20 minutes of TMB color development in the dark, and stop reaction with measurement at 450 nm. Serum and plasma sample preparation calls for centrifugation at 1000 x g for 20 or 15 minutes respectively, with tissue homogenate, cell culture supernatant and cell lysate protocols also described. Storage is 2-8°C for 12 months, with the microplate and lyophilized standard additionally suitable for 12 months at -20°C.
