FCHSD2 (F-BAR and double SH3 domains protein 2) is a multi-domain scaffold featuring an N-terminal F-BAR module that binds curved phosphatidylinositol-rich membranes and twin C-terminal SH3 domains that recruit dynamin, N-WASP, and other proline-rich ligands to sites of actin polymerization. The Assay Genie human FCHSD2 immunoassay provides a quantitative colorimetric readout for measuring endogenous FCHSD2 protein levels in research samples, supporting investigation of this scaffold in cytoskeletal and membrane-trafficking studies.

This scaffold couples membrane curvature sensing to branched actin assembly, contributing to clathrin-mediated endocytosis, membrane tubulation, lamellipodial protrusion, and polarized cargo trafficking. Documented research applications include studies of neuronal vesicle transport in hippocampal neurons, GLUT4 vesicle mobilization in adipocytes and myotubes downstream of insulin signaling, immunological synapse assembly in lymphocytes, and altered expression of membrane-remodeling proteins in migrating cancer cells.

The kit ships as 96 Assays on a pre-coated microplate, supplied with lyophilized recombinant standards, biotinylated detection antibody, streptavidin-HRP conjugate, wash buffer concentrate, TMB substrate solution, and stop reagent. The workflow involves sequential incubation, wash, and color-development steps compatible with standard plate readers. Researchers working with cell lysates, clarified tissue homogenates, and clarified research specimens can integrate the kit into ELISA workflows for research use only. Components should be stored refrigerated and protected from light until first use; reconstituted reagents are typically aliquoted and frozen for repeated runs, with repeated freeze-thaw cycles avoided to preserve binding activity.
