The target analyte, FCGR3B (also known as CD16b), is a glycosylphosphatidylinositol (GPI)-anchored low-affinity Fc gamma receptor expressed predominantly on the surface of human neutrophils, where it engages IgG-opsonized particles and immune complexes. Through this binding it contributes to antibody-dependent cellular cytotoxicity, phagocytosis, and neutrophil activation. The protein is highly polymorphic: the neutrophil antigen NA1 and NA2 allotypes differ in glycosylation and functional activity, and FCGR3B copy number variation has been associated with susceptibility to autoimmune conditions including systemic lupus erythematosus and granulomatosis with polyangiitis. The mature glycoprotein migrates at approximately 50-65 kDa under reducing conditions. Researchers use this in vitro ELISA kit from Assay Genie to quantify soluble or shed receptor material across multiple biological specimens, supporting studies in immunology, transfusion medicine, and autoimmunity biomarker discovery. The assay employs a human-specific capture antibody with no significant cross-reactivity to homologous Fc receptors, ensuring accurate measurement of FCGR3B. The microplate kit ships with pre-coated wells, lyophilized standards, biotinylated detection antibody, and color development reagents.
