Description

A sandwich ELISA, double-antibody kit from Assay Genie for the quantitative measurement of human FAM151A in serum, plasma, cell culture supernatant, cell or tissue lysate and other liquid samples. Detection is colorimetric, using a biotin-labelled detection antibody and an HRP-streptavidin (SABC) signal generation step with TMB substrate.

Reported analytical performance includes a sensitivity of 46.875 pg/mL and a standard curve range of 78.125–5000 pg/mL. Linearity in spiked samples (n = 5) across 1:2, 1:4 and 1:8 dilutions spanned 84–101% for serum, 82–105% for EDTA plasma and 82–105% for heparin plasma. Spike-and-recovery averaged 100% from serum (range 93–105%), 99% from EDTA plasma (range 91–104%) and 97% from heparin plasma (range 88–104%). The microplate is read at 450 nm after the TMB reaction is stopped.

FAM151A (family with sequence similarity 151 member A), also referenced as UNQ3034, PRO9836 or C1orf179 (UniProt Q8WW52), is a protein linked to cell growth, development and metabolism, and its dysregulation has been associated with several diseases, making it a useful biomarker for related research. Kit reagents are stored at 2–8°C for 12 months, with light-sensitive components kept away from direct light. The supplied protocol runs from reagent and plate equilibration through a 37°C primary incubation of 90 minutes, biotin-labelled detection antibody binding for 60 minutes, HRP-streptavidin binding for 30 minutes and TMB colour development in the dark for 10–20 minutes before absorbance measurement.
