Quantitative measurement of soluble FAS receptor in human samples is enabled by this Assay Genie capture immunoassay, formatted for use on standard microplate readers. The kit targets the death receptor also known as CD95, APO-1, or TNFRSF6, supporting research into apoptosis, immune cell turnover, autoimmune lymphoproliferative syndrome, tumor immune evasion, and graft-versus-host disease. Validated sample inputs include clinical specimens, tissue homogenates, cell lysates, and cell culture supernatants.

FAS is a type I transmembrane death receptor belonging to the TNF receptor superfamily. Engagement by FAS ligand (FASLG, CD95L) recruits FADD and procaspase-8 to form the death-inducing signaling complex, activating caspase-8 and the extrinsic apoptotic cascade. Soluble FAS (sFAS) isoforms generated by alternative mRNA splicing can antagonize FASLG binding, and elevated circulating sFAS has been reported in systemic lupus erythematosus, rheumatoid arthritis, certain leukemias, and solid tumor contexts. Quantifying sFAS therefore provides a window into both pro-apoptotic signaling and its dysregulation in disease.

The kit includes a pre-coated microplate, biotinylated detection reagent, streptavidin-HRP conjugate, reference standards, and assay buffer sufficient for 96 determinations. Reagents should be refrigerated upon arrival and equilibrated to room temperature before use. After addition of substrate and stop solution, absorbance is measured, with results calculated against the standard curve. The assay is intended for research use only and is not validated for diagnostic procedures.
