Human FABP5/E-FABP ELISA Kit

Assay Genie's human FABP5/E-FABP ELISA kit is a sandwich ELISA, double antibody colorimetric assay intended for the quantitative measurement of FABP5 (also known as E-FABP, a fatty acid-binding protein involved in lipid homeostasis and cell signalling). The kit is validated for use with human serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, and is associated with Uniprot Q01469 and the Signal Transduction research area.

The assay delivers a sensitivity of 0.188 ng/mL and a working range of 0.313-20 ng/mL. Reagent storage is at 2-8°C for 12 months. The six-step workflow begins with reagent and TMB substrate equilibration to room temperature; standards, samples and a zero control are then loaded onto the pre-coated plate for a 90 minute primary incubation at 37°C. A biotin-labelled detection antibody is added and incubated for 60 minutes, HRP-Streptavidin (SABC) for 30 minutes, and TMB substrate in the dark for 10-20 minutes. The reaction is then stopped and absorbance read at 450 nm.

Sample preparation guidance is provided for serum (clot then centrifuge at 1000 × g for 20 minutes), plasma (anticoagulant tubes, centrifuge at 1000 × g for 15 minutes at 2-8°C), tissue homogenate, cell culture supernatant (centrifuge 2500 rpm for 5 minutes), and cell lysate. Reported linearity across 1:2, 1:4 and 1:8 dilutions is 88-104% for serum (n=5), 82-101% for EDTA plasma (n=5), and 81-101% for heparin plasma (n=5). Average recovery is 99% for serum, 93% for EDTA plasma, and 95% for heparin plasma.
