Description

This Sandwich ELISA kit from Assay Genie is designed for the quantitative measurement of human F5 (Factor V) in research samples. Factor V, also known as coagulation Factor V, is a key protein in the blood coagulation cascade, and its dysregulation has been implicated in clotting disorders such as the Factor V Leiden mutation, which is associated with an increased risk of thrombosis. The kit uses a double-antibody sandwich format with a colorimetric detection method and is suited to laboratories researching hemostasis, thrombosis, and cardiovascular coagulation disorders.

Performance specifications include a sensitivity of 9.375 pg/mL and a standard range of 15.625–1000 pg/mL. In linearity testing, EDTA plasma samples (n=5) gave 87–97% at 1:2, 82–98% at 1:4, and 83–95% at 1:8, while heparin plasma samples (n=5) gave 91–104%, 87–100%, and 84–94% at the same dilutions. Spike-and-recovery testing returned ranges of 85–103% (average 95%) for EDTA plasma and 89–104% (average 96%) for heparin plasma.

The kit accepts plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, with serum-compatible preparation guidance also described. Components are stored at 2–8°C for 12 months, and several reagents can additionally be kept for 1 month at 2–8°C followed by 12 months at –20°C. The protocol involves a 90-minute primary incubation at 37°C, a 60-minute biotin-labelled detection antibody step, a 30-minute HRP-Streptavidin (SABC) binding step, TMB colour development in the dark for 10–20 minutes, and absorbance reading at 450 nm after stop solution addition.
