ESRRB is an orphan nuclear receptor (NR3B2) that occupies a distinctive niche in stem cell biology: it sustains naive pluripotency in pre-implantation embryos and embryonic stem cells by reinforcing the OCT4-SOX2-NANOG network at self-renewal gene enhancers. The receptor is therefore a frequent biomarker in regenerative medicine studies, reprogramming workflows aimed at producing naive induced pluripotent stem cells, and investigations of early developmental signaling. Aberrant ESRRB expression has also been documented in hormone-responsive tumors, including breast, prostate, and endometrial cancers, supporting its use as a quantitative endpoint in oncology research.
