Description

A colorimetric sandwich ELISA from Assay Genie for the quantitative detection of human ERGIC-53 (LMAN1) in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit uses a double-antibody sandwich format and is positioned for research in cell biology. ERGIC-53, also known as LMAN1, is a mannose-specific lectin involved in protein trafficking and glycosylation within the ER-Golgi intermediate compartment, and the target has been described as a valuable biomarker for studying conditions such as congenital disorders and neurodegenerative diseases, with elevated levels associated with certain cancers, autoimmune diseases, and genetic disorders.

The assay has a stated sensitivity of 46.875 pg/mL and a working range of 78.125-5000 pg/mL. Reagents should be stored at 2-8°C for up to 12 months. The protocol includes reagent and plate preparation, primary incubation at 37°C for 90 minutes, biotinylated detection antibody binding at 37°C for 60 minutes, HRP-streptavidin binding at 37°C for 30 minutes, TMB color development in the dark for 10-20 minutes, and a stop step with absorbance read at 450 nm. Reported linearity across 1:2, 1:4, and 1:8 dilutions spans 86-100% for serum, 83-100% for EDTA plasma, and 80-95% for heparin plasma, with spike-and-recovery averages of 99% in serum, 93% in EDTA plasma, and 94% in heparin plasma.
