Description

From Assay Genie, this sandwich ELISA kit is designed for the quantitative measurement of ERCC1 (Excision Repair Cross-Complementation Group 1) in human samples. The assay uses a double-antibody sandwich format with colorimetric detection and is suitable for use with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. ERCC1 is a key protein involved in DNA repair and the maintenance of genomic stability, and its dysregulation has been linked to cancer and neurodegenerative disorders, making it a relevant biomarker for research in disease progression and therapeutic response.

Linearity across dilutions of 1:2, 1:4 and 1:8 was 93–104%, 92–101% and 93–100% for serum (n=5); 82–92%, 84–97% and 82–98% for EDTA plasma (n=5); and 87–95%, 81–98% and 93–100% for heparin plasma (n=5). Spike-and-recovery averages were 94% for serum, 91% for EDTA plasma and 93% for heparin plasma, with corresponding recovery ranges of 85–103%, 86–98% and 87–100%.

The procedure involves equilibrating reagents, incubating standards and samples at 37°C for 90 minutes, adding biotin-labelled detection antibody for 60 minutes, then HRP-Streptavidin for 30 minutes, followed by TMB substrate development in the dark for 10–20 minutes, and reading absorbance at 450 nm after adding stop solution. Components should be stored at 2–8°C, with the kit remaining stable for 12 months under these conditions.
