Description

Assay Genie provides a sandwich ELISA, double antibody kit for the quantitative colorimetric detection of human Estrogen Receptor alpha (ER alpha / NR3A1) in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The assay delivers a sensitivity of 0.375 ng/mL across a working range of 0.625-40 ng/mL, and is intended for research into estrogen receptor alpha biology.

Estrogen receptor alpha, also known as NR3A1, is a key regulator of gene expression in response to estrogen signaling and plays a role in cell growth, differentiation, and development; its dysregulation has been implicated in diseases such as breast cancer and osteoporosis. The sandwich ELISA workflow includes equilibration of reagents and TMB substrate, primary incubation at 37°C for 90 minutes, addition of a biotin-labelled detection antibody with incubation at 37°C for 60 minutes, HRP-Streptavidin (SABC) binding for 30 minutes at 37°C, TMB color development in the dark for 10–20 minutes, and absorbance reading at 450 nm after stop solution. The kit is stored at 2-8°C for 12 months.

Linearity has been demonstrated across serum, EDTA plasma, and heparin plasma at 1:2, 1:4, and 1:8 dilutions, with serum ranges of 87-101%, 85-102%, and 88-99%, EDTA plasma ranges of 86-100%, 85-100%, and 86-98%, and heparin plasma ranges of 86-99%, 86-99%, and 80-89%. Recovery is 87-104% (average 96%) in serum, 86-102% (average 93%) in EDTA plasma, and 89-99% (average 94%) in heparin plasma.
