Description

Designed for quantitative measurement of EpCAM (Epithelial Cell Adhesion Molecule) in human samples, this Sandwich ELISA kit from Assay Genie uses a double-antibody, colorimetric detection workflow with absorbance read at 450 nm. The assay is intended for use with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid sample types.

Reported sensitivity is 46.875 pg/mL, with a working range of 78.125–5000 pg/mL (1IU=50pg). Linearity (n=5) has been demonstrated across 1:2, 1:4 and 1:8 dilutions: serum 89–104% / 86–101% / 83–98%, EDTA plasma 89–102% / 82–101% / 85–98%, and heparin plasma 87–101% / 82–101% / 87–100%. Recovery ranges (n=5) were 87–100% (average 94%) for serum, 85–105% (average 94%) for EDTA plasma, and 93–102% (average 96%) for heparin plasma. The kit should be stored at 2–8°C for 12 months.

The protocol proceeds through reagent and TMB substrate equilibration to room temperature, primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody binding at 37°C for 60 minutes, HRP-Streptavidin (SABC) incubation at 37°C for 30 minutes, TMB colour development in the dark for 10–20 minutes, and a stop reaction with immediate absorbance measurement at 450 nm. Sample preparation guidance includes clotting and centrifugation at 1000 × g for 20 minutes for serum, anticoagulant collection with centrifugation at 1000 × g for 15 minutes at 2–8°C for plasma, homogenisation in PBS with protease inhibitors for tissue, centrifugation at 2500 rpm for 5 minutes for cell culture supernatant, and lysis buffer with protease inhibitors for cell lysates.
