Human ENPP-7/Alk-SMase ELISA Kit

A Sandwich ELISA from Assay Genie for the colorimetric detection of ENPP-7 (also known as alkaline sphingomyelinase, or Alk-SMase) in human samples, this kit supports research into lipid metabolism and inflammation regulation, where ENPP-7 dysregulation has been linked to neuropsychiatric disorders, diabetes, and cancer.

The assay is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, with a sensitivity of 0.188 ng/mL and a working range of 0.313–20 ng/mL. Components are stored at 2–8°C for 12 months.

Linearity across 1:2, 1:4, and 1:8 dilutions was 86–102%, 92–104%, and 89–103% for serum; 85–99%, 87–96%, and 85–100% for EDTA plasma; and 85–100%, 82–93%, and 85–96% for heparin plasma. Spike-and-recovery experiments averaged 92% for serum (range 85–103%), 99% for EDTA plasma (range 85–104%), and 98% for heparin plasma (range 89–104%). The protocol involves antigen binding at 37°C for 90 minutes, biotin-labeled detection antibody incubation at 37°C for 60 minutes, HRP-streptavidin binding at 37°C for 30 minutes, TMB substrate development in the dark for 10–20 minutes, and absorbance measurement at 450 nm.
