Human Engrailed-2/EN2 ELISA Kit

A colorimetric Sandwich ELISA Kit from Assay Genie for the quantitative measurement of human Engrailed-2 (EN2) in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The double-antibody sandwich format uses a biotin-labeled detection antibody and HRP-Streptavidin conjugate with TMB substrate, and absorbance is read at 450 nm. Reported sensitivity is 0.188 ng/mL with a working range of 0.313-20 ng/mL. In spike-and-recovery testing (n=5 per matrix), serum averaged 95% (range 85-102%), EDTA plasma averaged 95% (range 87-100%), and heparin plasma averaged 94% (range 89-100%). Linearity at 1:2, 1:4 and 1:8 dilutions was 88-98%/86-98%/88-98% for serum, 86-102%/87-101%/80-90% for EDTA plasma, and 88-104%/84-101%/80-99% for heparin plasma. The protocol comprises a 90-minute primary incubation at 37°C, a 60-minute detection antibody step, a 30-minute HRP-Streptavidin step and 10–20 minutes of TMB colour development in the dark. Supplied components include a pre-coated dismountable microplate, lyophilized standard, biotin-labeled antibody, HRP-Streptavidin conjugate, TMB substrate, dilution buffers, wash buffer, stop solution and plate sealer; the kit is stored at 2-8°C for 12 months.

Engrailed-2 is a critical protein that plays a key role in development, particularly in patterning the midbrain and cerebellum. Dysregulation of EN2 has been linked to autism spectrum disorder and schizophrenia, making it a valuable biomarker for studying these conditions and potential therapeutic interventions.
