Beyond the basics described above, this Assay Genie EG-VEGF kit is configured as a complete sandwich immunoassay with a typical runtime of roughly 3 to 4.5 hours at room temperature, depending on whether a 1 hour or overnight 16 hour sample incubation is selected. The pre-coated 96-well microplate is supplied in a removable 8-well strip frame held in a resealable pouch with desiccant, so a partial plate can be run without wasting unused wells; once the pouch is opened, the strips remain stable for several months when kept refrigerated at 2-8 °C and protected from moisture.

Reported performance characteristics include a standard curve spanning approximately 31.25 to 2000 pg/mL, a limit of detection in the low double-digit pg/mL range, intra-assay precision below 8% CV and inter-assay precision below 10% CV, and spike-recovery generally between 85 and 115% across validated serum and EDTA plasma matrices. Each well takes a 100 µL sample, lyophilized calibrators are reconstituted in 1 mL of standard diluent, and the colorimetric reaction is developed with TMB substrate, stopped with acid, and read at 450 nm with optional 570 nm or 630 nm wavelength correction to subtract plate background.

EG-VEGF, also referenced by the gene symbols PROK1 and MIT1, is a secreted ligand for the G-protein-coupled receptors PROKR1 and PROKR2, so it is studied both as a tissue-selective angiogenic factor and as a GPCR signaling molecule. Published applications include preeclampsia biomarker surveys, follicular fluid and granulosa cell profiling in polycystic ovary syndrome, placental trophoblast invasion and decidualization assays, testicular Leydig and Sertoli cell function, adrenal cortex vascular remodeling, and tumor microenvironment studies of steroid-producing tissues. Compatible matrices in the published literature include serum, heparin plasma, EDTA plasma, citrate plasma, conditioned cell culture medium, and clarified tissue homogenates; grossly hemolyzed, icteric, or lipemic specimens should be avoided or interpreted with caution, and reconstituted standards should be aliquoted and frozen to limit repeated freeze-thaw cycles.
