In the sandwich-ELISA workflow, standards or samples are added to wells pre-coated with an antibody specific to the target protein, followed by a biotinylated detection antibody and an Avidin-Horseradish Peroxidase (HRP) conjugate. Free components are washed away and a substrate solution is added; wells containing the target develop a blue colour that turns yellow upon addition of stop solution. Absorbance is measured at 450 nm ± 2 nm and is directly proportional to the concentration of the target protein as determined from a standard curve. Kit components are stored at 2-8°C.
