Description

From Assay Genie, this colorimetric sandwich ELISA uses a double-antibody format with a biotinylated detection antibody and an HRP-Streptavidin (SABC) signal system. The TMB substrate produces a blue colour that turns yellow after stop solution is added, and absorbance is read at 450 nm using a microplate reader. The assay is designed for the quantitative measurement of human extracellular superoxide dismutase (EC-SOD/SOD3) and is validated for use with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples of human origin.

The kit reports a sensitivity of 0.375 ng/mL and an assay range of 0.625-40 ng/mL. Dilution linearity across serum, EDTA plasma, and heparin plasma (n=5) falls within 80-105% at 1:2, 1:4, and 1:8 dilutions. Spike recovery ranges from 85-105% for serum (average 96%), 86-104% for EDTA plasma (average 97%), and 89-103% for heparin plasma (average 97%). Reagents are stored at 2-8°C, with the included Bradford Reagent held at 4°C for total protein quantification and sample normalisation. The protocol uses incubations of 90 minutes for primary antigen binding at 37°C, 60 minutes for the biotinylated detection antibody, and 30 minutes for HRP-Streptavidin, with TMB colour development in the dark for 10-20 minutes before stop solution and absorbance reading.

For research use only, the kit is shipped on ice packs with a 12-month shelf life when stored at 2-8°C. Components include a pre-coated 96-well microplate, lyophilised standard, biotin-labelled detection antibody concentrate (100X), HRP-Streptavidin conjugate (SABC, 100X), TMB substrate, sample and antibody dilution buffers, SABC dilution buffer, wash buffer (25X), stop solution, plate sealer, technical manual, and a validated Bradford Reagent.
