Description

From Assay Genie, this competitive ELISA kit is designed for the quantitative detection of human E3 ubiquitin-protein ligase UBR4 in human samples, including serum, plasma, tissue homogenates, cell culture supernates and other biological fluids. UBR4 is a key component of the N-end rule pathway, recognising and binding to proteins bearing destabilising N-terminal residues and targeting them for ubiquitination and degradation. Dysregulation of UBR4 has been implicated in cancer, neurodegenerative disorders and autoimmune diseases, supporting its value as a biomarker for research into disease mechanisms and potential therapeutic interventions.

The kit provides a detection range of 0.625-40ng/mL with a sensitivity of 0.22ng/mL. Intra-assay CV is 5.2% and inter-assay CV is 8.3%. Linearity at 1:2 to 1:16 dilutions (N=5) spans 103-113% to 90-90% in serum, 109-117% to 107-119% in EDTA plasma, and 86-96% to 110-120% in heparin plasma. Average recovery is 97% in serum (range 91-103%) and 99% in plasma (range 93-105%). Specificity covers natural and recombinant human E3 ubiquitin-protein ligase UBR4.

Alternative names for UBR4 include 600 kDa retinoblastoma protein-associated factor, N-recognin-4, RING-type E3 ubiquitin transferase UBR4, Retinoblastoma-associated factor of 600 kDa, Zinc finger UBR1-type protein 1, RBAF600, p600, KIAA0462 and KIAA1307. The protein is a 574 kDa multi-pass membrane component found in membrane, cytoplasm, cytoskeleton and nucleus (UniProt Q5T4S7), with its gene located at chromosomal position 1p36.13. UBR4 interacts with RB1 and calmodulin, and is a target of the E7 oncoprotein from human papillomavirus HPV-16, HPV-6B and HPV-11. The kit is for research use only.
