Description

This competitive ELISA kit from Assay Genie is designed for the quantitative measurement of DYN/Big Dynorphin in human samples. It uses a competitive ELISA format coated with antigen and a colorimetric detection method, providing a sensitivity of 4.688 pg/mL and a working range of 7.813–500 pg/mL. The kit is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid sample types, with storage recommended at 2–8°C in a sealed state; cryopreservation should be avoided.

Validated linearity and recovery data demonstrate reliable performance. In linearity testing with serum, EDTA plasma, and heparin plasma (n = 10) at 1:2, 1:4, and 1:8 dilutions, results ranged from 80–104% across matrices. Recovery testing yielded average recoveries of 92% for serum (range 86–105%), 93% for EDTA plasma (range 87–102%), and 91% for heparin plasma (range 85–99%).

The six-step assay procedure involves equilibrating TMB substrate for 30 minutes at room temperature, adding 50 µL of standard or sample followed by 50 µL of biotin-labeled antibody per well and incubating at 37°C for 45 minutes, washing the plate five times with a one-minute soak, developing color with TMB substrate in the dark at 37°C for 10–20 minutes, adding stop solution (color change from blue to yellow), and reading absorbance at 450 nm. Dynorphin is a neuropeptide that modulates pain perception, mood, and addiction, and its dysregulation has been implicated in various neurological and psychiatric disorders, supporting research applications in pain management, mood disorders, and addiction.
