Human DUSP4/MKP2 ELISA Kit

This Sandwich ELISA kit from Assay Genie is designed for the quantitative measurement of DUSP4 (also known as dual-specificity phosphatase 4 or MKP-2) in human samples. It uses a double-antibody sandwich format with a colorimetric readout and supports research into cell signalling pathways governing cell growth, differentiation and the response to stress.

The assay is reactive with human samples and accepts serum, plasma, cell culture supernatant, cell or tissue lysates, and other liquid sample types. Reported performance includes a sensitivity of 0.188 ng/mL and a detection range of 0.313–20 ng/mL. Linearity has been characterised with n = 5 per matrix: serum 85–97% (1:2), 83–100% (1:4), 82–93% (1:8); EDTA plasma 88–98% (1:2), 82–101% (1:4), 87–93% (1:8); heparin plasma 91–105% (1:2), 82–100% (1:4), 81–95% (1:8). Spike-and-recovery averages are 99% for serum (range 93–103%), 98% for EDTA plasma (range 93–101%) and 93% for heparin plasma (range 88–98%).

The procedure uses a TMB substrate with absorbance measured at 450 nm after a stop step, and the kit is stored at 2–8°C for 12 months. DUSP4/MKP-2 negatively regulates MAP kinases (ERK1, ERK2 and JNK) and dysregulation of the phosphatase has been linked to cancer, inflammatory disorders and neurological conditions, making it a useful biomarker for studies of disease mechanisms and therapeutic targets.
