Human DPP9/Dipeptidyl peptidase 9 ELISA Kit

From Assay Genie, this Sandwich ELISA, Double Antibody kit is designed for the precise measurement of Dipeptidyl Peptidase 9 (DPP9) levels in human samples including serum, plasma, cell culture supernatants, cell or tissue lysates, and other liquid samples. DPP9 is an enzyme involved in regulating biological processes including immune response and inflammation, and its dysregulation has been linked to conditions such as autoimmune diseases and cancer.

The kit reports a sensitivity of 0.469 ng/mL and a range of 0.781-50 ng/mL, with detection carried out by colorimetric measurement at 450 nm after TMB substrate development. Linearity in serum (n=5) was 95-101%, 89-101% and 87-100% at 1:2, 1:4 and 1:8 dilutions; EDTA plasma (n=5) 87-99%, 82-97% and 82-98%; Heparin plasma (n=5) 88-98%, 83-95% and 85-99%. Recovery averaged 96% for serum, 93% for EDTA plasma and 95% for Heparin plasma, with corresponding ranges of 85-102%, 86-105% and 89-99%.

The assay protocol proceeds through reagent and plate preparation, a primary incubation at 37°C for 90 minutes, biotin-labeled detection antibody binding at 37°C for 60 minutes, HRP-Streptavidin binding at 37°C for 30 minutes, TMB color development in the dark for 10-20 minutes, and a stop reaction read at 450 nm. Sample preparation guidance is provided for serum, plasma, tissue homogenate, cell culture supernatant, cell lysate and other sample types. Components include a dismountable ELISA microplate, lyophilized standard, 100X biotin-labeled antibody, 100X HRP-Streptavidin conjugate (SABC), TMB substrate, sample dilution buffer, antibody dilution buffer, SABC dilution buffer, stop solution, 25X wash buffer, plate sealer and a technical manual. Storage is at 2-8°C for 12 months, with lyophilized standards and strips also stable at -20°C for 12 months.
