Description

A colorimetric Sandwich ELISA from Assay Genie for the quantitative measurement of human DPP10 (Inactive Dipeptidyl Peptidase 10) in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit employs a double-antibody sandwich format using a biotin-labeled detection antibody and HRP-Streptavidin (SABC) binding, with absorbance read at 450 nm following TMB color development.

Reported performance includes a sensitivity of 0.188 ng/mL and an assay range of 0.313-20 ng/mL. Linearity in serum (n=5) spans 88-104% at 1:2, 84-101% at 1:4, and 89-95% at 1:8, with comparable ranges observed for EDTA plasma (85-97%, 84-100%, 87-98%) and heparin plasma (87-102%, 84-101%, 80-100%). Recovery averages 98% in serum (range 86-104), 97% in EDTA plasma (range 92-101), and 93% in heparin plasma (range 89-101). Unopened kit storage is at 2-8°C for 12 months.

The assay procedure involves equilibrating reagents and TMB substrate to room temperature, loading standards and samples onto the pre-coated plate and incubating at 37°C for 90 minutes, followed by biotin-labeled detection antibody incubation at 37°C for 60 minutes, HRP-Streptavidin (SABC) binding for 30 minutes, TMB color development in the dark for 10-20 minutes, and absorbance reading at 450 nm after stop solution. Sample preparation guidance is provided for serum, plasma, tissue homogenate, cell culture supernatant, and cell lysate. DPP10 is described as an enzyme involved in protein metabolism and immune response regulation, with dysregulation linked to autoimmune diseases, neurological disorders, and cancer. Associated synonyms include DPL2, DPPY, DPRP3, and DPRP-3 (UniProt Q8N608).
