The DPAGT1 enzyme, also known as Dolichyl-Phosphate N-Acetylglucosaminephosphotransferase 1, is an integral endoplasmic reticulum membrane protein that catalyzes the first committed step of N-linked protein glycosylation by transferring GlcNAc-1-P from UDP-GlcNAc onto dolichyl phosphate. Mutations in the DPAGT1 gene are linked to congenital disorder of glycosylation type Ij (CDG-Ij) and to autosomal recessive limb-girdle congenital myasthenic syndromes, while altered expression has been reported in hepatocellular carcinoma and T-cell lymphoma models, making accurate quantitation useful for glycobiology, neurology, and oncology research teams.

This Assay Genie kit is supplied as a 96-assay plate with capture antibody pre-coated onto the wells, along with biotinylated detection antibody, protein standard, streptavidin-HRP conjugate, and TMB substrate; each well is loaded with 100 µL of sample. The plate is processed through incubation, wash, and chromogenic development steps, with absorbance read on a standard plate reader. To preserve reagent activity, reconstituted vials should be aliquoted and stored at -20°C with minimal freeze-thaw cycles, and the unopened plate should be kept refrigerated at the conditions listed on the kit insert until first use.
