The Assay Genie Human DOHH (Deoxyhypusine hydroxylase) ELISA Kit enables species-specific quantitative measurement of human DOHH, the iron/2-oxoglutarate-dependent dioxygenase that catalyzes the final hydroxylation step in hypusine biosynthesis on eukaryotic translation initiation factor 5A (eIF5A).

DOHH is a 302-residue enzyme that converts deoxyhypusine on eIF5A to mature hypusine-eIF5A, a modification essential for proper translation of mRNAs containing polyproline stretches and for nuclear export of Rev-containing HIV-1 transcripts. Because the hypusine pathway is the only known source of this residue, DOHH activity is a bottleneck for eIF5A-dependent processes, and altered DOHH expression has been documented in proliferative diseases, certain solid tumors, and viral replication models.

The kit is supplied as a complete microplate-based immunoassay with pre-coated plates, lyophilized standards, detection antibody, and substrate reagents sufficient for one full plate run. Researchers should store reagents refrigerated at 4 °C and equilibrate plate components to room temperature before running the assay to ensure reproducible binding kinetics.

Typical research uses include profiling DOHH expression in patient cohorts, biomarker discovery in oncology, characterization of eIF5A pathway inhibitors such as GC7 and ciclopirox, and investigation of viral-host interactions. The platform is configured for in vitro research use only.
