DNA (cytosine-5)-methyltransferase 3A sits at the center of de novo methylation in human cells, where it establishes new methyl marks on CpG dinucleotides during development, X-inactivation, and tumor suppression. The enzyme is mutated in a substantial fraction of adult acute myeloid leukemia cases and underlies Tatton-Brown-Rahman syndrome, making accurate protein quantification a recurring need across hematology, developmental biology, and epigenetics research programs.

This quantitative ELISA from Assay Genie is supplied as a 96-assay kit intended for in vitro research use only. It is configured to detect the human DNMT3A protein across sample matrices that include conditioned media from cultured cells and peripheral blood fractions, using an absorbance-based readout. Reagents should be stored refrigerated at 4 degrees C upon arrival and protected from light; standards and detection antibody are provided in individual vials, while the assay plate is supplied as breakable strip wells for flexible throughput.

Typical applications include monitoring DNMT3A expression in leukemia cell lines before and after treatment with hypomethylating agents such as azacitidine or decitabine, profiling primary patient samples alongside mutational screening, and characterizing isogenic gene-edited systems in which DNMT3A has been knocked out or catalytically inactivated. The kit also supports studies of DNMT3A regulation by histone marks, transcription factors, and DNMT3L, and pairs naturally with methylation-specific PCR or bisulfite sequencing to connect protein levels with downstream methylation outcomes.

Samples should be aliquoted and frozen at -80 degrees C where possible to minimize freeze-thaw cycles, since repeated thawing degrades DNMT3A recovery from complex matrices. Standard curves are constructed from recombinant human DNMT3A provided with each kit, and absorbance values are recorded on a standard microplate reader fitted with the appropriate filter. A full protocol insert covering dilution recommendations for cell culture supernatant and blood-derived specimens is supplied by Assay Genie with every shipment.
