Human DNAH17 ELISA Kit

A sandwich, double-antibody ELISA from Assay Genie for the quantitative measurement of human DNAH17 (Dynein Axonemal Heavy Chain 17), a component of cilia and flagella involved in cell motility, with dysregulation linked to primary ciliary dyskinesia and infertility. Detection is colorimetric, using a TMB substrate read at 450 nm.

The kit reports a sensitivity of 18.75 pg/mL and a working range of 31.25-2000 pg/mL. Validated sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Reported linearity values for serum (n=5) are 88-100% at 1:2, 86-96% at 1:4, and 80-92% at 1:8; EDTA plasma shows 91-102% / 85-101% / 87-97% across the same dilutions, and heparin plasma 90-103% / 90-101% / 82-99%. Spike-and-recovery averages are 96% for serum, 93% for EDTA plasma, and 93% for heparin plasma. The assay procedure involves a 90-minute primary incubation at 37°C, a 60-minute biotinylated detection antibody binding step, a 30-minute HRP-streptavidin (SABC) incubation, and 10-20 minutes of TMB development in the dark, with all reagents stored at 2-8°C for 12 months.

Sample preparation guidance includes clotting and centrifuging serum at 1000 × g for 20 minutes, collecting plasma from anticoagulant tubes centrifuged at 1000 × g for 15 minutes at 2-8°C, clarifying cell culture supernatant at 2500 rpm for 5 minutes, and lysing cells in buffer containing protease inhibitors. The kit is supplied with a dismountable pre-coated microplate, lyophilized standard, biotin-labelled antibody concentrate, HRP-streptavidin conjugate (SABC), TMB substrate, sample, antibody and SABC dilution buffers, stop solution, 25X wash buffer, plate sealer, and a technical manual.
